Quantity: 50μl/vial
Form: Liquid IgG fraction, affinity purified on immunogenic peptide column. The DTTpretreated
antibody was conjugated with succinimidyl 4-[N-maleimidomethyl]-cyclohexane-1-carboxylate (SMCC)-activated R-Phycoerythrin (RPE) under optimum conditions. The solution is free of unreacted SMCC-phycoerythrin.Stabilized with 2.5 mg/ml bovine serum albumin and Proclin 300 (0.1% v/v).
Immunogen: Synthetic peptide (15-mer peptide corresponding to aa 1242-1256 of C4) specific for human complement split product C4d coupled to KLH (keyhole limpet hemocyanine). The antibody was generated according to a previously published protocol (Ref 1,2).
Specificity: The antibody identifies human complement split product C4d. It specifically reacts with the C4d domain of C4 and therefore also binds to C4 andother C4dcontaining split products. A detailed characterization of thepolyclonal anti-C4d reagent is published (Ref 1,2).
Application: This antibody is applicable for detection of cell- or solid-phase-bound C4 and C4d
split product by flow cytometry and can be monitored in FL2 channel analyses on FACS devices. The conjugate has been especially established for Luminex®-based detection of alloantibody-triggered C4d deposition to HLA-coated microparticles (One Lambda, Inc., Canoga Park, CA, USA) according to a modification of [C4d]FlowPRA® testing (Ref 3-5).
